تأثیر فرمولاسیون و شرایط فرایند اکستروژن بر ویژگیهای میانوعده حاوی پودر عناب
مصرف بالای میانوعده کمارزش و سوءتغذیه اقشار جامعه بهخصوص کودکان و نوجوانان یکی از معضلاتی است که امروزه در جامعه وجود دارد. لذا در این پژوهش تولید میانوعده با ارزش غذایی بالا حاوی آرد عناب موردبررسی قرار گرفت. در این پژوهش بر پایه طرح مرکب مرکزی چرخشپذیر، اثر متغیرهای فرایند اکستروژن شامل سرعت چرخش ماردون (120-150-180 دور بر دقیقه)، افزودن آرد عناب (5-10-15 درصد) و رطوبت خوراک ورودی (12-16-20 درصد) بر ویژگیهای فیزیکوشیمیایی، بافتی و رنگ فراورده حجیمشده بر پایه ذرت-گندم موردبررسی قرار گرفت. نتایج نشان داد؛ افزودن آرد عناب تا 15 درصد بهطور مستقل موجب افزایش دانسیته 33/0، سختی بافت 73/35 نیوتن، حلالیت 42درصد و پارامتر قرمزی رنگ 41/3 و کاهش پارامتر روشنایی رنگ 1/89 شد. بااینحال اثر همزمان دو پارامتر سرعت چرخش و افزودن آرد عناب سبب بهبود خصوصیات فراورده گردید، بهنحوی که با افزایش سرعت چرخش ماردون و درنتیجه کاهش ویسکوزیته موجب کاهش سختی بافت تا 79/1 نیوتن گردید. شرایط بهینه تولید میانوعده اکسترودشده، شامل میزان رطوبت خوراک 19/13 درصد، میزان آرد عناب 30/6 درصد و سرعت چرخش ماردون 2/122 دور بر دقیقه تعیین گردید.

مطالعه خصوصیات فیزیکوشیمیایی آرد سه واریته کینوا (Quinoa) و بررسی اثر pH بر ویژگیهای عملکردی آن
حبوبات و غلات بهعلت پراکندگی کشت و گستردگی مصرف، بخش مهمی از غذای انسان را تشکیل میدهند، اما در این بین فقط چند خانواده خاص با اختلاف در سراسر جهان مصرف میشوند. یکی از این دانهها گندم است که با ویژگیهای منحصربهفرد خود در سازگاری کشت و خواص عملکردی، سهم بهسزایی از سفره جوامع بشری بهخود اختصاص داده است.از سال 2011 میلادی کینوا بهعنوان یکی از مهمترین شبهغلات در دنیا مطرح شد، بهطوریکه سال 2013 را بهعنوان سال بینالمللی کینوا نامگذاری نمودند، این دانه که حائز ویژگیهای منحصربهفردی در کشت و عملآوری است در سراسر جهان موردتوجه قرار گرفته است. از مهمترین ویژگیهای مطلوب این دانه خواص شیمیایی و عملکردی آن در مقایسه با دانههای مشابه است. در این پژوهش ضمن اندازهگیری خواص شیمیایی سه واریته پر کاربرد کینوا (سفید، قرمز و سیاه)، نتایج حاصل، با خواص دانههای مشابهی همچون عدس، نخود، جو، گندم، برنج، گاودانه، لوبیا سودانی و شنبلیله نیز مقایسه شدهاند که مشخصاً میزان چربی در واریته قرمز، میزان کربوهیدرات و خاکستر در واریته سفید و میزان رطوبت و چربی در واریته سیاه بیش از دو واریته دیگر بود. در مقایسه با سایر دانهها نیز چربی در کینوا مشابه گندم و بیش از برنج و جو بود. در زمینه خواص عملکردی نیز باید اشاره نمود که کمینه میزان حلالیت پروتئین در 5/4 pH= (که نقطه ایزوالکتریک پروتئینهاست) و بیشینه این مقدار، در 9 pH= مشاهده گردید. همچنین محاسبات نشان داد جذب آب و روغن در آرد این دانه بسیار بالا است. بیشینه مقدار مشاهدهشده در اندازهگیری کفکنندگی و پایداری آن، در 6pH= و واریته سفید بود و حداکثر پایداری نیز در زمان 30 دقیقه ثبت شد. درانتها نیز نتایج حاصل از بررسی خاصیت امولسیونکنندگی و پایداری امولسیون نشان داد، بیشینه این ویژگی در 9 pH=در واریته سفید و بهینهترین زمان پایداری نیز، 30 دقیقه در همین واریته است.

امروزه با تغییر فرهنگ زندگی، بروز سوءتغذیه در گروههای مختلف سنی، و بروز بیماریهایی نظیر دیابت، فشارخون و چاقی، تولید فرمولاسیونهای غذایی کمکالری امری ضروری بهنظر میرسد. از طرف دیگر، با افزایش تقاضا برای مصرف محصولات با ارزش غذایی و کیفیت بالا، استفاده از ترکیبات با منشأ طبیعی جهت ارتقاء ویژگیهای تغذیهای و کیفی محصولات غذایی توصیه میگردد. لذا، هدف از پژوهش حاضر، کاهش ساکارز مصرفی در تهیه لوز رژیمی و تولید محصولی با ارزش غذایی و اقتصادی بالا با افزایش جایگزینی گیاه آزیوش با پسته است. در این پژوهش اثر جایگزینی استویا با ساکارز (صفر-100 درصد)، جایگزینی آزیوش با پودر پسته (صفر-100 درصد) و دمای پخت (70-50 درجه سانتیگراد) بر دانسیته و سفتی لوز رژیمی موردبررسی قرار گرفت. روش سطح پاسخ در قالب طرح مرکب مرکزی با 6 نقطه مرکزی و دو تکرار و آلفای برابر با 2 در سایر نقاط بهمنظور بهینهیابی فرمولاسیون لوز رژیمی غنیشده، استفاده شد. ویژگیهای فیزیکوشیمیایی، بافت و رنگ در بازه زمانی 45 روزه برای نمونههای بهینه و شاهد مقایسه گردید. دانسیته تیمارها با افزایش درصد جایگزینی استویا و آزیوش افزایش یافت. اثر متقابل دو متغیر آزیوش و دمای پخت باعث کاهش سفتی بافت گردید. کاهش درصد جایگزینی استویا سفتی بافت را کاهش داد. فرمولاسیون بهینه لوز رژیمی شامل 25 درصد استویا، 25 درصد آزیوش با دمای پخت 65 درجه سانتیگراد؛ و 55 درصد استویا، 75 درصد آزیوش با دمای پخت 55 درجه سانتیگراد معرفی گردید. درمجموع، ارزش غذایی لوز رژیمی حاوی گیاه آزیوش از جنبه محتوی پروتئین، خاکستر، منیزیم و کلسیم بهبود یافت.

مدلسازی و بررسی تغییرات فیزیکوشیمیایی ماهی تونا تحتتأثیر روش انجمادزدایی اهمی غوطهوری
انجمادزدایی در مبحث صنایع غذایی همواره چالشی بزرگ بوده است، زیرا روشهای سنتی باعث کاهش کیفیت ماده غذایی میشوند. انجمادزدایی اهمی یک روش حرارتی جایگزین، سریع و همگن است. در این پژوهش، انجمادزدایی ماهی تونا با روش اهمی بهصورت غوطهوری در آبنمک با سه غلظت مختلف (3/0 ،4/0 و 5/0 درصد وزنی/حجمی سدیم کلرید) در شیب ولتاژ 143/7 ولت بر سانتیمتر و فرکانس 50 هرتز انجام شد. نمونهها تا 48 ساعت پس از انجمادزدایی در دمای 4 درجه سانتیگراد نگهداری شدند. تغییرات افت انجمادزدایی، افت پخت، افت کل، ظرفیت نگهداری آب، بافت، متغیرهای رنگی L*، a*، b* و تغییرات رنگ بهصورت مدلهای کینتیکی بررسی شدند. طبق نتایج، در طی زمان نگهداری افت انجمادزدایی، افت پخت، افت کل و ظرفیت نگهداری آب افزایش یافت. تغییرات رنگ (a* و b*) و بافت نیز با افزایش زمان و غلظت آبنمک در سطح احتمال 1% معنیدار شد. میزان تیوباربیتوریک اسید در اثر هیچ یک از متغیرهای مستقل معنیدار نشد. همچنین با افزایش غلظت آب نمک، افت پخت کاهش یافت. میزان نیروی برشی با گذشت زمان و افزایش غلظت آب نمک، کاهش یافت. متغیرهای وابسته با مدلهای کینتیکی مناسب با بالاترین ضریب رگرسیون توصیف شدند. همچنین مدلهای کینتیکی با R2 و RMSE اعتبارسنجی شدند. این نتایج نشاندهنده بهبود خصوصیات فیزیکی محصول و عدم تشدید اکسیداسیون با روش انجمادزدایی اهمی از طریق غوطهوری در آبنمک است.

بررسی اثر صمغ کنجاک، صمغ دانه مرو، سرعت همگنسازی و چربی بر pH، بافت و رنگ ماست همزده کمچرب
در این تحقیق اثر مقادیر مختلف صمغ کنجاک (صفر تا 2/0 درصد)، صمغ دانه مرو (صفر تا 2/0 درصد) بهعنوان متغیرهای آزمایشی طرح مخلوط و میزان چربی (صفر تا 3 درصد) و سرعت همگنسازی (صفر تا 24000 دور در دقیقه) بهعنوان متغیرهای آزمایشی طرح فرآیندی بر pH، بافت و رنگ ماست کمچرب با استفاده از طرح اپتیمال متقاطع مخلوط-فرایند موردبررسی قرار گرفت. نتایج بررسی pH نشان داد که فقط اثر چربی برpH نمونهها معنیدار بود، بهطوریکه با افزایش میزان چربی، pH نمونهها نیز افزایش یافت. آزمون اکسترژن برگشتی-آنالیز پروفیل بافت نشان داد که با افزایش میزان چربی و سرعت همگنسازی، سختی، نیروی چسبندگی، پیوستگی و حالت صمغی نمونهها بهطور معنیداری افزایش یافت. همچنین اگرچه در سرعتهای همگنسازی پایین دو صمغ کنجاک و دانه مرو بر سختی، نیروی چسبندگی، پیوستگی و حالت صمغی نمونهها اثر سینرژیستی داشتند، اما نتایج، نشاندهنده عملکرد بیشتر صمغ دانه مرو نسبت به صمغ کنجاک بر روی این خصوصیات در سرعت همگنسازی بالا بود. نتایج حاصل از اندازهگیری رنگ نشان داد که تنها اثر خطی صمغ دانه مرو و اثرات متقابل صمغ کنجاک-صمغ دانه مرو، صمغ کنجاک-سرعت هموژن بر روی *L نمونهها معنیدار بود و با افزایش میزان صمغ کنجاک و کاهش صمغ دانه مرو، *L نمونهها کاهش ولی با افزایش سرعت هموژن *L افزایش یافت. بهمنظور بهینهسازی صفات در این تحقیق، چربی و نیروی چسبندگی حداقل وسختی و پیوستگی حداکثر درنظر گرفته شدند که با توجه به صفات مذکور، میزان صمغ کنجاک 146/0 درصد، میزان صمغ دانه مرو 053/0 درصد، چربی 42/2 درصد و سرعت همگنسازی 12300 دور در دقیقه بهدست آمد.

بررسی اثر افزودن آرد قسمتهای مختلف سنجد (Elaeagnus angustifolia) بر ویژگیهای کیک اسفنجی بدون گلوتن
بیماری سلیاک نوعی بیماری خودایمنی گوارشی است که در اثر مصرف پروتئین گلوتن ایجاد میشود و تنها راه درمان این بیماری، استفاده از رژیم غذایی بدون گلوتن میباشد. امروزه بهواسطه توجه و علاقه مصرفکنندگان به ویژگیهای تغذیهای مواد غذایی درخواست برای تولید مواد غذایی فراسودمند افزایش یافته، پس برنامهریزی برای تولید فرآوردههای بدون گلوتن و همچنین بهبود کیفیت آنها بااهمیت است. میوه گیاه سنجد حاوی طیف وسیعی از ترکیبات غذایی مختلف و مفید و دارای خواص درمانی است که از آرد آن میتوان بهعنوان یک ماده اولیه عملگرا در تولید محصولات نانوایی برای تولید محصولی باکیفیت و ارزش تغذیهای بهتر و ایجاد تنوع در محصولات استفاده نمود. در این پژوهش، اثر افزودن آرد سنجد بهعنوان منبع فیبر و املاح معدنی، بهشکل آرد هسته سنجد، آرد پوسته و گوشت سنجد و آرد سنجد کامل هر کدام در سه سطح (5، 10 و 20 درصد بر اساس وزن آرد و یک نمونه شاهد) بر خصوصیات فیزیکوشیمیایی و حسی کیک اسفنجی بدون گلوتن بر پایه آرد ذرت بررسی شد. نتایج نشان داد میزان رطوبت، خاکستر، فیبر و مواد معدنی نمونهها افزایش و میزان چربی، پروتئین، pH و حجم مخصوص نمونههای کیک با افزایش درصد آردهای سنجد کاهش یافت. پارامترهای بافت (سفتی، پیوستگی و صمغی شدن) در روزهای 1، 7 و 14 روز پس از پخت افزایش سفتی و صمغی شدن و کاهش پیوستگی را نشان داد و اثر آردهای سنجد بر شاخصهای رنگی (L*، a*، b* و BI) معنیدار بود. بیشترین امتیاز ارزیابی حسی را نمونههای حاوی 5 و 20 درصد آرد پوسته و گوشت سنجد کسب کردند.

بررسی اثر روش استخراج با حلالهای آب و متانول بر ویژگیهای ضداکسایشی و ضدمیکروبی عصاره گیاه بنسرخ: مطالعه در شرایط آزمایشگاهی “in vitro”
در این پژوهش، عصاره گیاه بنسرخ با استفاده از حلالهای متانول و آب و بهکمک روش ماسراسیون استخراج شد. راندمان استخراج، ترکیبات فنول کل، فعالیت آنتیاکسیدانی (بر پایه مهار رادیکالهای 2 و 2-دیفنیل 1-پیکریل هیدرازیل (DPPH) و 2،2-آزینو بیس-3-اتیل بنزوتیازولین-6-سولفونیک اسید (ABTS)) و اثر ضدمیکروبی (بر پایه آزمونهای دیسک دیفیوژن آگار، حداقل غلظت مهارکنندگی و حداقل غلظت کشندگی) عصارهها با یکدیگر مقایسه گردید. راندمان استخراج و میزان ترکیبات فنولی عصاره متانولی بهترتیب 10/7 درصد و mg GAE/g 28/88 بود، درحالیکه برای عصاره آبی بهترتیب 6/4 درصد و mg GAE/g 29/68 بود. فعالیت آنتیاکسیدانی عصاره آبی (برحسب رادیکالهای DPPH و ABTS بهترتیب mg/mL 28/2 و 98/2) بهطور قابلتوجهی بالاتر از عصاره متانولی (برحسب رادیکالهای DPPH و ABTS بهترتیب mg/mL 33/5 و 12/7) بود که نشان میدهد وجود ترکیبات فنولی تنها عامل مؤثر بر ظرفیت آنتیاکسیدانی عصاره نمیباشد. میزان حداقل غلظت مهارکنندگی از رشد عصاره متانولی برای باکتریهای Streptococcus pyogenes، Listeria innocua، Entrobacter aerogenes، Escherichia coli و Pseudomonas aeruginosa بهترتیب 68/4، 68/4، 75، 75/18 و 75/18 میلیگرم بر میلیلیتر بود، درحالیکه برای عصاره آبی بنسرخ بهترتیب 68/4، 37/9، 75، 5/37 و 5/37 میلیگرم بر میلیلیتر بود. میزان حداقل غلظت کشندگی عصاره متانولی برای باکتریهای Streptococcus pyogenes، Listeria innocua، Entrobacter aerogenes، Escherichia coli و Pseudomonas aeruginosa بهترتیب 75/18، 75/18، 150، 150 و 5/37 میلیگرم بر میلیلیتر بود، درحالیکه برای عصاره آبی بنسرخ بهترتیب 5/37، 5/37، 300، 150 و 75 میلیگرم بر میلیلیتر بود. با توجه به نتایج، عصاره گیاه بنسرخ قابلیت استفاده بهعنوان نگهدارنده طبیعی جهت جلوگیری از اکسیداسیون غذاهای حاوی اسیدهای چرب غیراشباع و همچنین کنترل رشد باکتریهای بیماریزا و مولد فساد را دارا میباشد.

بررسی تأثیر افزودن کدو سبز (Cucurbita pepo) بر ویژگیهای فیزیکی-شیمیایی و حسی ماست همزده طی نگهداری
ماست از محصولات لبنی است که اثرات سلامتبخشی مانند کنترل مشکلات گوارشی، تحریک سیستم ایمنی و افزایش طول عمر دارد. فیبرها با جذب آب، باعث افزایش حجم غذا و تسهیل دفع میشوند. همچنین با افزایش مصرف فیبر، خطر بیماریهای قلبی و عروقی، چاقی مفرط، دیابت و سرطان روده بزرگ کاهش مییابد. کدوسبز از سبزیجاتی است که ویژگیهایی مفید مانند اثر بر سطح قند و چربی خون دارد. در این تحقیق اثر افزودن کدوسبز بهصورت پودر و گرانول در سطحهای 7/0، 4/1 و 2 درصد به ماست همزده بر ویژگیهای فیزیکی-شیمیایی شامل pH، اسیدیته، آباندازی، سفتی بافت، رنگ و ویژگیهای حسی موردبررسی قرار گرفت. همچنین اثر زمان نگهداری طی 1، 5، 10 و 15 روز بر این ویژگیها ارزیابی گردید. نتایج، بیانگر اثر معنیدار نوع، درصد کدوسبز و مدت زمان نگهداری بر pH، اسیدیته و سفتی بافت بود (P<0.05). طی زمان، اسیدیته، افزایش و pH کاهش یافت. در هیچیک از تیمارها در طول زمان، آباندازی مشاهده نشد. سفتی بافت نمونههای حاوی پودر کدوسبز بیشتر از نمونههای حاوی گرانول بود و طی زمان، سفتی بافت افزایش یافت. از روز اول تا روز پنجم، سفتی بافت افزایش یافت و طی روز پنجم به دهم کاهش سفتی مشاهده شد و نهایتاً تا روز پانزدهم سفتی بافت افزایش یافت. اثر نوع، سطح افزودن و اثر متقابل این متغیرها روی تغییرات شاخصهای L* و a* و b* رنگ معنیدار بود (P<0.05). با افزایش درصد کدو شاخص L* ابتدا روند افزایشی و سپس کاهشی داشت و شاخصهای a* و b* ابتدا روند کاهشی و سپس افزایشی داشتند. روشنی و سبزی رنگ نمونههای حاوی گرانول بیشتر از نمونههای حاوی پودر بود. مقدار شاخص a* تقریباً در نمونههای حاوی گرانول بیشتر از نمونههای حاوی پودر بود، درحالیکه مقدار شاخص b* در نمونههای حاوی پودر بیشتر بود. ویژگیهای حسی نمونهها (رنگ، طعم، رایحه، قوام، احساس دهانی و پذیرش کلی) بهطور معنیداری تحتتأثیر زمان افزایش یافت (P<0.05). ارزیابها تفاوتی در نوع و درصد کدوسبز قائل نشدند و بیشترین امتیاز شاخصهای حسی در روز پانزدهم بود و نمونه مطلوب ازنظر ارزیابها، ماست حاوی 4/1 درصد گرانول کدوسبز بود.

ارزیابی و مقایسه خصوصیات حسی، بافتی و رنگ خامه کمچرب با استفاده از تحلیل مؤلفه اصلی و رگرسیون حداقل مربعات جزئی
در این تحقیق اثر مقادیر مختلف صمغ کنجاک (2/0 تا 6/0 درصد)، میزان چربی (18 تا 25 درصد) و فشار هموژن (100 تا 200 بار) بر خصوصیات حسی، بافتی و رنگ خامه کمچرب موردبررسی قرار گرفت و سپس این خصوصیات با استفاده از روشهای تحلیل مؤلفه اصلی و رگرسیون حداقل مربعات جزئی مقایسه گردید. نتایج حاصل از ارزیابی ویژگیهای اندازهگیریشده با استفاده از روش سطح پاسخ حاکی از آن بود که افزایش فشار هموژن سبب افزایش سختی، قوام، چسبندگی، *b، امتیاز حسی بافت، امتیاز حسی عطر و پذیرش کلی نمونهها شد، درحالیکه افزایش چربی منجر به افزایش چسبندگی، *L، *b، امتیاز حسی طعم، امتیاز حسی عطر و پذیرش کلی نمونهها گردید. افزایش صمغ کنجاک نیز سبب افزایش سختی، قوام، چسبندگی و امتیاز حسی بافت شد. پراکنش پاسخها در فضای مؤلفههای اصلی نیز نشان داد که صفات دستگاهی سختی و قوام در مجاورت هم و نزدیک به امتیاز حسی بافت قرار گرفتند که مجاورت این صفات حاکی از آن بود که صفات اندازهگیریشده توسط ارزیابها تقریباً معادل پارامترهایی بود که توسط روشهای دستگاهی اندازهگیری شده بودند. همچنین نتایج حاصل از ارزیابی ضریب همبستگی میان دادههای حسی و دستگاهی نشان داد که بالاترین میزان همبستگی بین سختی و امتیاز حسی بافت (یک ارتباط خطی مثبت قوی (774/0)) و قوام و امتیاز حسی بافت ( یک ارتباط خطی مثبت قوی (760/0)) وجود داشت.

بررسی آثار تغییرات دمایی بر پارامترهای سینتیکی-اکسایشی تریآسیلگلیسرولهای زیتون، کانولا و کنجد در حضور آنتیاکسیدان سزامول
در این پژوهش شاخصهای سینتیکی-اکسایشی تریآسیلگلیسرولهای زیتون، کنجد و کانولا در حضور سطوح غلظتی 01/0 تا 16/0 درصد سزامول و دماهای 60، 80 و 100 درجه سانتیگراد موردبررسی قرار گرفتند. نتایج حاکی از آن بود که افزایش دما عدد پراکسید متناظر با طول دوره اکسایش کند (PVIP) را برای تریآسیلگلیسرولهای کنجد و کانولا کاهش داده است، لیکن تغییرات این شاخص برای تریآسیلگلیسرولهای زیتون بیمعنی بود. ضرایب حرارتی بهدست آمده در کلیه غلظتهای سزامول برای تریآسیلگلیسرولهای روغن زیتون نسبت به دو روغن دیگر کمتر بود که نمایانگر پایداری حرارتی بیشتر این روغن در حضور این آنتیاکسیدان است. ثابت سرعت مصرف سزامول (kS) برای تریآسیلگلیسرولهای زیتون، کنجد، و کانولا در غلظت 01/0 درصد و دمای 60 درجه سانتیگراد بهترتیب برابر با 72/1، 92/1، و h-1 34/2 بود که با افزایش دما بهشکل محسوسی این ثابت افزایش یافت، بهطوریکه روند تغییرات این شاخص برای سامانههای تریآسیلگلیسرولی غیراشباعتر بیشتر بود.

تولید بستنی مبتنی بر تخممرغ فراسودمند حاوی ایمونوگلوبین Y بر علیه باکتری هلیکوباکتر پیلوری
امروزه، مصرفکنندگان بهدنبال غذاهایی هستند که باعث سلامتی شده و خطر ابتلا به بیماریها را کاهش دهد. تخممرغ بهعنوان منبع بزرگی از ایمونوگلوبین Y است که نقش بسیار مهمی در ایمنی غیرفعال دارد. ازاینرو، این پژوهش با هدف ایجاد ایمنی غیرفعال خوراکی در برابر باکتری هلیکوباکتر پیلوری که شیوع بسیار وسیعی در جهان داشته و عامل اصلی زخم و سرطان معده است، صورت پذیرفت. بدین منظور، سلولهای باکتری هلیکوباکتر پیلوری با استفاده از فرمالدهید غیرفعال و پس از ترکیب با ادجوانت فعال و غیرفعال فروند به 4 نقطه از سینه مرغ نژاد لگهورن سفید تزریق زیرجلدی شد. استخراج آنتیبادی IgY با استفاده از پلیاتیلین گلایکول و کیفیت و غلظت آن با استفاده از روشهای SDS-PAGE و برادفورد موردبررسی قرار گرفت. از زرده تخممرغهای حاوی IgY بهمنظور تولید بستنی تخممرغی استفاده و محصول در مقایسه با بستنی وانیلی مورد ارزیابی حسی قرار گرفت. از تست الایزا غیرمستقیم بهمنظور بررسی فعالیت IgY استخراجی، همچنین فعالیت IgY در طی سه ماه پس از تولید استفاده شد. حضور دو باند 25 و 67 کیلودالتونی بر روی ژل نشان از کیفیت مناسب روش استخراج داشت و غلظت IgY استخراجی 46/11 میلیگرم بر میلیلیتر برآورد شد. آنالیز فعالیت IgY با روش الایزا نشان داد که بیشترین میزان این آنتیبادی در هفته دوم داشته و بعد از 3 ماه نگهداری بستنی تخممرغی در دمای 18-، 94 درصد از فعالیت آنتیبادی حفظ شده است. بررسی ارزیابی حسی بین بستنی حاوی IgY و نمونه شاهد نشان دهنده عدم تفاوت معنی داردر 5 پارامتر اصلی تست چند نقطه ای می باشد. بنابراین، با توجه به فعال بودن آنتیبادی IgY در آزمایشهای الایزا، امکان اتصال IgY حاضر در بستنی تخممرغی تولیدشده در این پژوهش با باکتری هلیکوباکتری پیلوری فراهم بوده و این محصول میتواند در پیشگیری از تکثیر باکتری هلیکوباکتری پیلوری مؤثر عمل کند.

بهینهسازی تولید نانوامولسیون اسانس پونه کوهی ایرانی حاوی اسیدهای چرب امگا سه با استفاده از روش امولسیفیکاسیون خودبهخودی
اخیراً استفاده از روشهای کمانرژی بهدلیل عدم نیاز به تجهیزات گرانقیمت و سهولت تولید بسیار رواج یافته است. در این پژوهش، نانوامولسیون اسیدهای چرب امگا سه با استفاده از اسانس پونه کوهی ایرانی بهروش امولسیفیکاسیون خودبهخودی تهیه گردید. هدف از این مطالعه، بررسی شرایط بهینه تولید نانوامولسیون با استفاده از طرح دیاپتیمال بود، برای این منظور نانوامولسیونهای حاوی اسیدهای چرب امگا سه با استفاده از اسانس گیاهی پونه کوهی ایرانی با چهار متغیر مستقل غلظت امگا سه (75-25 درصد)، نوع سورفاکتانت (توئین 80، توئین 20، توئین 80:20 و کازئینات سدیم)، نسبت سورفاکتانت به روغن (300-10 درصد) و مدت زمان نگهداری (60-1 روز) تولید گردیدند و تأثیر آنها بر پارامترهایی مانند میانگین قطر ذرات، توزیع اندازه ذرات، شاخص کدورت، اندیس خامهای شدن، خواص آنتیاکسیدانی، اندیس پراکسید، اندیس تیوباربیتوریک اسید، ضریب شکست، بریکس و pH موردمطالعه قرار گرفت و سپس بهمنظور تعیین بهترین فرمولاسیون بهینهسازی انجام گرفت. نتایج نشان دادند که میانگین اندازه قطرات بسیار تحتتأثیر غلظت سورفاکتانت و نوع سورفاکتانت بود و با افزایش مقدار سورفاکتانت، میانگین قطر قطرات کاهش معنیداری یافت، درواقع افزایش غلظت سورفاکتانت در نانوامولسیونها در اکثر موارد سبب افزایش میزان جذب سورفاکتانت به سطح W/O گردیده و درنتیجه منجر به کاهش بیشتر کشش سطحی شد و درنهایت سبب کاهش اندازه ذرات و کاهش اندیس خامهای شدن گردید، همچنین افزایش غلظت سورفاکتانت، بهعلت کاهش میانگین قطر ذرات، پراکنش نوری کاهش یافته و میزان کدورت کاهش یافت، از طرفی افزایش غلظت سورفاکتانت و کاهش اندازه ذرات، در کاهش اندیس پراکسید نیز مؤثر بود. با استفاده از بهینهسازی عددی مقادیر بهینه متغیرهای مستقل نانوامولسیونهای پونه کوهی ایرانی حامل اسیدهای چرب امگا سه بهترتیب، میزان امگا سه 31/29 درصد، مدت زمان نگهداری 40 روز، درصد SOR (100) درصد و نوع سورفاکتانت توئین 80:20 تعیین شدند.

قهوه از یکی از مهمترین و پرطرفدارترین نوشیدنیها در دنیا بهشمار میرود، بهطوریکه از لحاظ تنوع روشهای تهیه بر سایر نوشیدنیهای مشابه مانند چای برتری دارد. در میان ترکیبات مختلف موجود در دانه قهوه، کافئین از اهمیت ویژهای برخوردار است. کافئین یک آلکالوئید طبیعی است که میتواند بسته به میزان مصرف، خواص مثبت (بهبود حافظه، افزایش هوشیاری و تمرکز) یا منفی (سردرد، اضطراب و بیقراری) بر سلامت مصرفکننده داشته باشد. از طرف دیگر، مقدار زیاد کافئین موجود در دانه قهوه بهعنوان یک عامل مزاحم در آنالیز آلایندهها عمل میکند و باید تا حد ممکن از عصاره حذف شود. ازاینرو پژوهش حاضر دو هدف کلی را دنبال کرد. در مرحله اول 18 نوع قهوه، اعم از انواع کلاسیک (جوشیده، فرانسوی، موکا و فیلترشده) و تجاری (انواع اسپرسو، قهوه فوری و قهوه سرد) از لحاظ میزان کافئین موردبررسی قرار گرفتند. در مرحله دوم، تأثیر دو روش (روش کچرز و متداول) بر میزان استخراج کافئین از پودر قهوه ربوستا، موردمطالعه قرار گرفت. نتایج این پژوهش مؤید تأثیر قابلملاحظه واریته بر میزان کافئین بود بهنحویکه قهوههای تهیهشده از دانه ربوستا کافئین بیشتری را نسبت به دانههای عربیکا دارا بودند. در میان انواع قهوه کلاسیک، بیشترین میزان کافئین (26±1044 میلیگرم بر لیتر) از قهوه موکا استخراج شد. در میان انواع تجاری، میزان قهوه به آب متفاوت بود لذا میزان کافئین برحسب مقدار دریافت از هر فنجان موردارزیابی قرار گرفت. در این میان، انواع اسپرسو، خصوصاً نوع پاد محتوی بیشترین میزان کافئین (16±1371 میلیگرم بر لیتر معادل 6/0±54 در هر فنجان 40 میلیلیتری) بودند. در مقابل کمترین میزان کافئین از قهوه سرد بهدست آمد. بررسی روش استخراج قهوه نیز نشان داد که روش کچرز قادر است کافئین را به شکل قابلملاحظهای (حدود 20% کاهش در مقایسه با روش متداول) از عصاره قهوه حذف کند که احتمالاً بر کاهش اثر مداخلهکننده این ترکیب در آنالیز آلایندهها که معمولاً در مقادیر بسیار کم در نمونهها وجود دارند، تأثیر مثبت خواهد داشت. بهطورکلی، استفاده از حلال استونیتریل و استفاده همزمان از جاذبهای PSA و C18، کاهش معنادار استخراج کافئین را بهدنبال داشت.

تولید و ارزیابی خصوصیات فیزیکوشیمیایی و رئولوژیکی شورتنینگ کماشباع با استفاده از تکنیک اورگانوژلسازی، مبتنی بر مونوآسیل گلیسرول و پلیگلیسرول استر
در این تحقیق، از تکنیک اورگانوژلسازی مبتنی بر مونوآسیل گلیسرول و پلیگلیسرول استر بهمنظور تولید شورتنینگ با میزان اشباعیت پایین استفاده شده است. بدینمنظور، یک گروه مخلوط دوتایی متشکل از 20% روغن پالم استئارین و80% روغن کلزا ساخته شد، بهطوریکه درصد اسیدهای چرب اشباع در ترکیب نهایی 20% گردید. اورگانوژلاتورهای مورداستفاده ترکیبی از مونوآسیل گلیسرول و پلیگلیسرول استر میباشند که در 6 نسبت دوتایی (0-100)، (20-80)، (40-60)، (60-40)، (80-20) و (100-0) مخلوط و در سه غلظت 3، 6 و 9 درصد (وزنی/وزنی) به ترکیب روغن اولیه افزوده شدند و خواص فیزیکوشیمیایی آنها مانند ترکیب اسیدهای چرب، نقطه ذوب لغزشی، درصد چربی جامد، عدد پراکسید، درصد اسید چرب آزاد و همچنین خواص رئولوژیکی شامل مدولهای ویسکوز، الاستیک، کمپلکس و زاویه فاز بررسی گردیدند. نتایج نشان داد که اسیدهای چرب اشباعشده، نقطه ذوب لغزشی، محتوای چربی جامد و مقاومت اکسایشی نمونهها با افزایش نسبت اولئوژلها از 3 به 9% افزایش یافته است. مقادیر عدد پراکسید و اسیدهای چرب آزاد اولئوژلهای مختلف تولیدشده که به روغن افزوده شدند، تفاوت معنیداری نشان نداد. در تمامی اولئوژلها، مقادیر مدول الاستیک از مدول ویسکوز بیشتر میباشد که نشان میدهد اولئوژلهای تولیدی دارای خواص شبهجامد بیشتری بودند. اولئوژلهای تهیهشده با نسبت 100 به صفر پلیگلیسرول استر و مونوآسیل گلیسرول دارای بیشترین مدول کمپلکس بودند. نتایج بررسی زاویه فاز اولئوژلها در تطابق با یافتههای مدول الاستیک بود و نسبتهای 100 به صفر و 20 به 80 پلیگلیسرول استر و مونوآسیل گلیسرول دارای مقاومت بالاتری بوده و نسبت به سایر نمونهها خواص مایع گونه را دیرتر ظاهر ساختند. بررسی پتانسیل کاربردی اولئوژلهای تولیدی مشخص نمود که آنها در تولید مارگارینهای نانوایی و شورتنینگهای چندمنظوره مناسب میباشند.

مقایسه اثرات صمغهای قدومه شیرازی و فارسی بهعنوان هیدروکلوئید جایگزین چربی بر خصوصیات فیزیکوشیمیایی و حسی خامه صبحانه کمچرب
این تحقیق با هدف جایگزینی بخشی از چربی خامه صبحانه با استفاده از دو نوع صمغ قدومه شیرازی و فارسی انجام شد. بدین منظور میزان چربی خامه از 25 درصد (در نمونه شاهد) به 18 و 15 درصد کاهش یافت و هریک از این صمغها بهطور جداگانه در مقادیر 2/0، 3/0 و 4/0 درصد (وزنی/وزنی) در فرمولاسیون استفاده شدند. سپس ویژگیهای فیزیکوشیمیایی و حسی نمونهها موردبررسی و مقایسه قرار گرفت. نتایج نشان داد که اثر نوع صمغ و مقدار صمغ روی میزان pH، اسیدیته، ماده خشک، رطوبت، آباندازی و ویسکوزیته نمونههای تولیدشده معنیدار بود (05/0>P). در هر دو نوع خامه (با 15 و 18 درصد چربی)، pH تیمار حاوی %4/0 صمغ قدومه شیرازی (81/6) بهطور معنیداری بالاتر از pH نمونه شاهد (69/6) و دیگر تیمارها بود (05/0>P). درصد رطوبت نمونه شاهد (%7/68) پایینتر از تیمارهای موردبررسی (%7/78-73) بود اما درصد ماده خشک نمونه شاهد (%3/31) بالاتر از تیمارها (%9/26-2/21) بود و بیشترین ماده خشک در هر دو نوع خامه 18 و 15 درصد به تیمار حاوی %4/0 صمغ فارسی اختصاص یافت. ویسکوزیته تیمارهای خامه %18 چربی حاوی %4/0 صمغ قدومه شیرازی (21915 سانتیپوآز) و صمغ فارسی (21919 سانتیپوآز) بهطور معنیداری بالاتر از نمونه شاهد (21507 سانتیپوآز) و سایر تیمارها بود. همچنین آباندازی در تیمارهای خامه %18 چربی حاوی %4/0 صمغ قدومه و صمغ فارسی (صفر میلیلیتر) و نیز تیمار خامه %15حاوی %4/0 صمغ قدومه شیرازی (صفر میلیلیتر) همانند نمونه شاهد مشاهده نگردید (05/0<P). در ویژگیهای حسی نیز نوع صمغ صرفاً روی رنگ اثر معنیدار داشت و اثر نوع خامه و مقدار صمغ عمدتاً روی بافت و پذیرش کلی نمونهها معنیدار بود. درمجموع خامه کمچرب %18 چربی حاوی %4/0 صمغ قدومه و صمغ فارسی بهترین نتایج فیزیکوشیمیایی و حسی را در مقایسه با سایر تیمارها بهخود اختصاص دادند.
