بهینهسازی تولید کف در خشککردن کفپوشی آبکرفس و ارزیابی خواص پودر تولیدی
در این پژوهش، تولید پودر آبکرفس با استفاده از روش خشککردن کفپوشی موردمطالعه قرار گرفت. شرایط تولید کف بهینه آبکرفس از نظر غلظت صمغ زانتان (45/0-25/0 درصد) بهعنوان پایدارکننده کف، مقدار کنسانتره پروتئین آبپنیر (6-2 درصد) بهعنوان عامل کفزا و زمان همزدن (10-2 دقیقه) جهت ایجاد کف با کمترین مقدار دانسیته و بیشترین پایداری با استفاده از روش سطح پاسخ تعیین شد. سپس کف بهینه تولیدی در سه دمای 40، 55 و 70 درجه سانتیگراد و ضخامت 3 میلیمتر خشک شد و پودرهای تولیدی از نظر رطوبت، فعالیت آبی، دانسیته توده، دانسیته ضربه، دانسیته ذره، تخلخل و میزان پیوستگی و جریانپذیری موردمطالعه قرار گرفتند. درنهایت مشخص شد که افزایش دمای خشککردن از 40 به 70 درجه سانتیگراد، تأثیر معناداری (05/0>p) بر ویژگیهای فیزیکوشیمیایی پودرها داشت و سبب کاهش میزان رطوبت، فعالیت آب، دانسیته توده، دانسیته ضربه، دانسیته ذره و نسبت هاسنر و شاخص کار شد ولی میزان تخلخل افزایش یافت.

ارزیابی ویژگیهای پروبیوتیکی و ضدباکتریایی SL163-4 Lactobacillus fermentum
پروبیوتیکها میکروارگانیسمهایی غیربیماریزایی هستند که از طریق تعادل میکروبی (نسبت باکتریهای مفید و بیماریزا) در روده انسان میتوانند باعث سلامتی و ایمنی گردند. این مطالعه با هدف بررسی پتانسیل پروبیوتیکی سویه Lactobacillus fermentum جداشده از غذای تخمیری انجام شد. در این پژوهش، ویژگیهای پروبیوتیکی Lactobacillus fermentum شامل: مقاومت به اسید (pH 5/2 ،5/3 و 5)، مقاومت به صفرا (2/0، 5/0، 8/0، 2/1 و 3 درصد)، فعالیت ضدمیکروبی (بهروش نقطهگذاری بر باکتریهای Listeria innocua، Staphylococcus aureus و Pseudomonas aeruginosa) و مقاومت نسبت به آنتیبیوتیکهای رایج درمانی (کلرامفنیکل، تتراسایکلین، پنیسیلین و جنتامایسین) بررسی شد. نتایج نشان داد که سویه Lactobacillus fermentum در 5/2=pH توانایی رشد را ندارد اما در 5/3=pH و 5/5=pH بهترتیب 92 و 99 درصد زندهمانی مشاهده شد. Lactobacillus fermentum توانست در تمامی غلظتهای نمکهای صفراوی رشد کند؛ هرچند با افزایش درصد غلظت نمک صفراوی، رشد باکتری کاهش یافت. میانگین قطر هاله عدم رشد برایListeria innocua ،Staphylococcus aureus و Pseudomonas aeruginosa بهترتیب 60/12، 20 و 10/11 میلیمتر تعیین شد. Lactobacillus fermentum نسبت به آنتیبیوتیکهای کلرامفنیکل، تتراسایکلین، پنیسیلین حساس و نسبت به جنتامایسین نیمهحساس بود (مقایسه با جداول CLSI). با توجه به نتایج پژوهش حاضر، Lactobacillus fermentum دارای قابلیت پروبیوتیکی قابلقبولی بود و میتوان از آن بهعنوان یک باکتری پروبیوتیک در محصولات غذایی بهره برد.

تأثیر کاربرد ایزوله پروتئین جوانه گندم بر ویژگیهای فیزیکی، رئولوژیکی، بافت وحسی بستنی
غنیسازی محصولات لبنی، خصوصاً بستنی جهت تولید فرآوردههای غذایی سلامتیبخش یکی از مهمترین اهداف و اولویتهای بشر امروزی میباشد. ایزوله پروتئینی یکی از ترکیبات مورداستفاده بهمنظور تولید محصولات سلامتیبخش است. در این مطالعه، هدف تولید بستنی با ویژگیهای جدید بر پایه یک ماده فراسودمند با جایگزینی جزئی ماده خشک با ایزوله پروتئین جوانه گندم میباشد. بنابراین ایزوله پروتئینی جوانه گندم با ترکیب درصد 1، 3 و 5 استخراج و به بستنی اضافه شد. در قدم اول ایزوله پروتئینی توسط آزمونهای درصد رطوبت، درصد چربی، خاکستر غیرمحلول در اسید، اندازه ذرات و دانهبندی آرد موردارزیابی قرار گرفت. در گام دوم ایزوله پروتئینی جوانه گندم با ترکیب درصد 1، 3 و 5 به بستنی اضافه شد و اثر آن بر روی ویژگیهای رئولوژیکی، خصوصیات بافتی، رنگ، پارامترهای فیزیکوشیمیایی و حسی بستنی موردارزیابی قرار گرفت. نتایج نشان داد که درصد رطوبت ایزوله پروتئینی و میزان درصد چربی بهترتیب عبارت بودند از 42/0±41/8 و 44/0±34/11 و همچنین میزان درصد خاکستر غیرمحلول در اسید عبارت بود از 23/0±57/3 . نتایج ارزیابی اندازه ذرات و دانهبندی آرد نیز عبارت بودند از 01/0±282. دادههای آزمایشگاهی نمایشگر آن بود که با افزایش ایزوله پروتئین جوانه، ویسکوزیته، مقاومت در برابر ذوب شدن و شاخص حجمافزایی بهطور معنیداری کاهش یافت. پارامترهای سختی و چسبندگی بافت، دانسیته ، اندازه و قطر ذرات و همچنین توزیع آنها و سطح مخصوص آنها، شاخصهای زردی و قرمزی بهطور معنیداری افزایش یافت. با جمعبندی کلیه نتایج آزمایشگاهی و ارزیابی حسی نمونهها مشخص شد که نمونه بستنی با یک درصد ایزوله پروتئینی جوانه گندم نمونه منتخب و بهینه میباشد.

تعیین ساختار، ترکیبات و ویژگیهای شیمیایی، فعالیت ضداکسایشی و اثر سیتوتوکسیک اسانس زردچوبه
هدف از این پژوهش شناسایی ترکیبات، ساختار و تشخیص کیفی نوع پیوندها، فعالیت ضداکسایشی، تعیین فنول و فلاونوئید کل و اثر سیتوتوکسیک اسانس زردچوبه بود. اسانس زردچوبه با دستگاه کلونجر و با روش تقطیر آبی استخراج شد. ترکیبات شیمیایی اسانس زردچوبه با دستگاه کروماتوگرافی گازی متصل به طیفسنج جرمی شناسایی شد. برای تعیین گروههای عاملی ترکیبات شیمیایی و شناسایی کیفی اسانس زردچوبه از آنالیز طیفسنجی تغییرشکل فروسرخ فوریه در محدوده طول موج cm-1 4000-500 استفاده شد. پتانسیل آنتیاکسیدانی اسانس زردچوبه با مهار رادیکالهای آزاد (DPPH و ABTS) و رنگبری بتاکاروتن لینولئیک اسید تعیین گردید. مقدار فنول کل و فلاونوئید با روشهای رنگسنجی اندازهگیری شد. از روش MTT جهت تعیین اثر سیتوتوکسیک غلظتهای مختلف اسانس زردچوبه بر رده سلولی سرطان روده بزرگ (HT29) استفاده شد. 18 ترکیب شناسایی شده در اسانس زردچوبه 91/97 درصد ترکیبات را تشکیل دادند. Turmerone با 40% بیشترین ترکیب شناسایی شده در اسانس زردچوبه بود. محدوده عدد موجی cm-1 3600-3400 (بهویژه عدد موجی cm-1 3516) و پیکهای cm-1 2930، 1621، 1515 و 1447 بهترتیب مربوط به ارتعاشات کششی پیوندهای O-H، C-H، C=O و C=C حلقه آروماتیک و گروههای آروماتیک کورکومینوئیدها است. پتانسیل آنتیاکسیدانی اسانس زردچوبه بهروشهای DPPH، ABTS و رنگبری بتاکاروتن لینولئیک اسید بهترتیب 15/25، 90/93 و 76/72 درصد بود. فنول کل و فلاونوئید کل اسانس زردچوبه بهترتیب mg GAE/g 91/38 و mg QE/g 9/87 بود. نتایج نشان داد که با افزایش غلظت اسانس زردچوبه از mg/mL 125/3 به 200 میزان زندهمانی سلول HT29 بهترتیب از 76/66 به 88/9 درصد تغییر پیدا کرد.

بهینهسازی فرمول بستنی وانیلی با کمک جایگزینی آرد کینوا با شیر خشک بدون چربی بهروش سطح پاسخ
کینوا بهعنوان یک ترکیب عملگرا، از نظر محتوی پروتئین، اسیدهای آمینه ضروری مانند لیزین و متیونین، اسیدهای چرب ضروری و اسیدهای چرب غیراشباع، مواد معدنی مانند آهن، کلسیم، روی، مس، فیبر رژیمی و ویتامینها و مواد آنتیاکسیدانی از اهمیت ویژهای برخوردار میباشد. در پژوهش حاضر از آرد کینوا با سطح جایگزینی با شیر خشک بدون چربی (صفر تا صد درصد)، روغن هیدروژنه گیاهی (5/4 تا 5/8 درصد) و صمغ پانیسول (25/0 تا 65/0 درصد) جهت دستیابی به فرمول بهینه تولید بستنی وانیلی استفاده گردید. بهینهسازی فرمولاسیون براساس پارامترهای تغییرات حجم، سرعت ذوب و خواص بافتی (سفتی و چسبندگی) توسط روش سطح پاسخ در قالب طرح مرکب مرکزی با 6 نقطه مرکزی و دو تکرار (2=α) در سایر نقاط صورت گرفت. نتایج نشان داد که برای روند تغییرات تغییرات حجم، مدل خطی پیشنهاد شد و اثر مستقل جایگزینی آرد کینوا، اثر متقابل جایگزینی آرد کینوا و سطوح مختلف صمغ پانیسول، اثر مجذور جایگزینی آرد کینوا، اثر مجذور سطوح مختلف روغن هیدروژنه گیاهی و اثر مجذور سطوح مختلف صمغ بر فاکتور تغییرات حجم معنیدار بود(05/0>P). جایگزینی آرد کینوا در فرمولاسیون بستنی، تأثیر قابلتوجهی بر میزان تغییرات حجم، سرعت ذوب و سفتی بافت داشت. فرمول بهینه پیشنهادی برای سطوح جایگزینی آرد کینوا با شیر خشک بدون چربی بهترتیب 25 و 53 درصد، روغن هیدروژنه گیاهی 5/8 و 2/8 درصد و صمغ پانیسول 39/0 و 48/0 درصد بود.

تهیه فیلم کامپوزیت زیستتخریبپذیر کربوکسیمتیل سلولز-صمغ عربی و ارزیابی خصوصیات فیزیکی، مکانیکی و حرارتی آن
در این پژوهش اثر ترکیب دو پلیمر کربوکسیمتیل سلولز و صمغ عربی بهمنظور بهبود خواص فیزیکی و مکانیکی فیلمهای زیستتخریبپذیر موردمطالعه قرار گرفت. بدین منظور ابتدا فیلمهای کربوکسیمتیل سلولز و صمغ عربی بهصورت جداگانه تهیه و در ادامه این دو پلیمر به نسبتهای مختلف (75:25، 50:50 و 25:75) با یکدیگر ترکیب شده و خصوصیات فیزیکی، مکانیکی و حرارتی آنها ارزیابی گردید. میزان نفوذپذیری به بخار آب در فیلمهای ترکیبی دو پلیمر با افزایش سطح صمغ عربی بهطور معنیداری کاهش یافت (05/0>p) و کمترین میزان عبور بخار آب در تیمار 75:25 (کربوکسیمتیل سلولز: صمغ عربی) 2567/2 (g pas-1 m-1 s-1) مشاهده شد. میزان آبگریزی فیلمها با اضافه کردن صمغ عربی به ماتریس فیلم کربوکسیمتیل سلولز بهطور معنیداری از 33/41 درجه به 10/61 درجه افزایش یافت (05/0>p). مقاومت کششی فیلمهای تولیدشده با افزایش سطح صمغ عربی از 10/51 مگاپاسکال به 16/3 مگاپاسکال کاهش یافت. میزان عبور نور از فیلمهای تولیدشده با افزایش نسبت صمغ عربی کاهش نشان داد (05/0>p). بیشترین و کمترین میزان دمای انتقال ذوب بهترتیب در تیمارهای کربوکسیمتیل سلولز (29/265 درجه سانتیگراد) و صمغ عربی (9/241 درجه سانتیگراد) مشاهده شد. بهطورکلی، از نتایج حاضر دریافت میشود که تهیه فیلمهای ترکیبی از کربوکسیمتیل سلولز و صمغ عربی باعث بهبود برخی از ویژگیهای فیلمهای تولیدشده ازقبیل خواص فیزیکی، حرارتی و میزان عبور نور گردید.

بهینهسازی خواص مکانیکی و نوری نانوکامپوزیتهای بر پایه پلیوینیل الکل بهمنظور بستهبندی مواد غذایی
در این پژوهش، برای بهبود ویژگیهای فیلمهای بر پایه پلیوینیل الکل (PVA) از دو نوع نانوذره دیاکسید تیتانیوم (TiO2) (۱و۲ درصد وزنی) و مونت موریلونیت (MMT) (۲و ۴ درصد وزنی) بهصورت توأم استفاده گردید و سپس تأثیر این دو نانوذره بر خواص مکانیکی (مدول یانگ و مقاومت کششی)، ویژگیهای رنگ (تفاوت رنگسنجی کل و ضریب سفیدی) و میزان عبور نور در فیلمها توسط روش سطح پاسخ (RSM) موردبررسی قرار گرفت. جهت تعیین مشخصات فیلمها از تکنیکهای مختلفی نظیر پراش اشعه ایکس و میکروسکوپ الکترونی روبشی استفاده شد. الگوهای پراش X نشان دادند که نانوذرات بهخوبی در ماتریس پلیمر پخش شدهاند و ریزساختار فیلمهای PVA/TiO2 و PVA/MMT از نوع لایهلایه میباشد. همچنین میکروگرافهای SEM توزیع خوبی از نانوذرات را در غلظت کم نشان دادند، درحالیکه در غلظتهای بالا تجمع نانوذرات مشاهده شد. در این پژوهش اثر خطی نانوذرات MMT و اثر متقابل TiO2 و MMT بر مقاومت کششی معنیدار بود (۰۵/۰>P)، درحالیکه اثر خطی، درجه دوم و اثر متقابل هر دو نانوذره بر مدول یانگ معنیدار بود (۰۱/۰>P) و در حالت کلی مقادیر بهینه TiO2 و MMT بهترتیب ۱% و ۴% برای خواص مکانیکی بهدست آمد. همچنین وجود هردو نانوذره در ماتریس پلیمر بر میزان عبور نور از فیلم و ΔE مؤثر و معنیدار بود (۰۱/۰>P). با بررسی سفیدی فیلمها فیلم نانوکامپوزیتی با ترکیب ۲% از TiO2 و ۴% از MMT از نظر ظاهری سفیدتر از سایر نمونهها و در حقیقت کدرتر گزارش شد. با بررسی نتایج مختلف آزمایشگاهی و تحلیل آنها با روش سطح پاسخ نمونه بهینه از نظر خواص مکانیکی و فیزیکی نمونهای با TiO2 ۵/۰% و MMT ۴% پیشنهاد شد.

افزایش ماندگاری قارچ تکمهای با اسانس خوشاریزه و بستهبندی نانوکامپوزیت پلی اتیلن-رس
در این پژوهش، ویژگیهای کمی و کیفی قارچ تکمهای در دمای 4 درجه سانتیگراد طی 35 روز نگهداری در بستهبندیهای پلیاتیلن و نانوکامپوزیت پلیاتیلن-رس 5 درصد در ترکیب با اسانس خوشاریزه در سه غلظت 0، 50 و 100 میکرولیتر در لیتر بررسی شد. ارزیابی صفاتی ازقبیل کاهش وزن، سفتی بافت، نشت الکترولیت و محتوی مالون دیآلدیید، فعالیت آنزیمهای آنتیاکسیدانی، جمعیت میکروبی، شاخص قهوهای شدن، درصد باز شدن کلاهکها و بازارپسندی کلی قارچهای تکمهای در بستهبندیها نشان داد که افزودن نانوذرات رس در بستر پلیاتیلن اثر معنیداری بر افزایش دوره نگهداری نسبت به بستهبندی پلیاتیلن داشت. از طرف دیگر استفاده از اسانس خوشاریزه در هر دو غلظت 50 و 100 میکرولیتر در لیتر اثرات بهبوددهنده در حفظ ویژگیهای قارچها در دوره نگهداری در هر دو نوع بستهبندی نشان داد، بااینوجود، غلظت 100 میکرولیتر در لیتر اسانس خوشاریزه تأثیر بیشتری داشت. در کل، بستهبندی قارچهای تکمهای در نانو کامپوزیت پلیاتیلن-رس 5 درصد و استفاده از پوشش خوراکی اسانس خوشاریزه با غلظت 100 میکرولیتر در لیتر برای افزایش دوره نگهداری این محصول همراه با حفظ کیفیت پس از برداشت و بازارپسندی پیشنهاد میگردد.

بررسی اثر پیشفرآوری فراصوت و آنزیم آلفا-آمیلاز بر تولید نشاسته متخلخل از نشاستههای گندم و ذرت و بررسی میزان توانایی جذب یون آهن
نشاستههای متخلخل به گروهی از نشاستههای فرآوریشده اطلاق میشوند که بهعنوان یک جاذب پرقدرت، ترکیبات حساس به نور، حرارت و اکسیداسیون را در خود جای میدهند و علاوهبر نقش حامل بهعنوان محافظ نیز محسوب میشوند. تولید نشاستههای متخلخل امروزه با بهکارگیری فناوریهای نوین مانند امواج فراصوت و آنزیمها روبهگسترش است. در این پژوهش نشاسته گندم و ذرت با توان 350 وات بهمدت 10 دقیقه طی فرآیند فراصوت قرارگرفتند. مراحل افزودن آنزیم آلفا-آمیلاز (آنزیم باکتریایی از باسیلوس سوبتیلیس) در غلظت 1/0 درصد بهصورت قبل، همزمان و پس از اعمال عملیات فراصوت صورت گرفت. به نشاستههای گندم و ذرت متخلخل تولیدشده یون آهن (آمونیوم سولفات آهن (II)) در غلظتهای ppm 40، 60 و 80 افزوده شد. نشاستههای متخلخل پس از تولید، بهوسیله میکروسکوپ الکترونی مشاهده و توانایی آنها در جذب آب و روغن موردبررسی قرار گرفت. سپس میزان یون آهن جذبشده و نوع پیوندهای شکلگرفته بین نشاستهها و یون آهن بهترتیب توسط دستگاه پلاسمای جفتشده القایی (ICP) و طیفسنج مادون قرمز (FTIR) تعیین شدند. در کلیه مراحل نشاسته ذرت و گندم با یکدیگر مقایسه شدند. تصاویر SEM نشان دادند که نشاستههای گندم و ذرت که ابتدا تحت عملیات فراصوت و سپس آنزیم قرار گرفتند (گروه اول) دارای شکل منظمتری از تخلخلها هستند. همچنین قابلیت جذب آب و روغن در نشاستههای گروه اول تفاوت معناداری با نشاستههای گروه دوم (عملیات همزمان فراصوت و آنزیم) و سوم (ابتدا عملیات آنزیمزنی و سپس فراصوت) ندارند. مقادیر موردآزمون در نشاسته ذرت در کلیه مراحل بالاتر از نشاسته گندم بود. جذب یون آهن در نشاستههای متخلخل نسبت به نشاستههای معمولی افزایش یافته و با افزایش غلظت یون آهن، میزان جذب آن توسط نشاستهها افزایش یافت. همچنین این نشاستهها قادر به تشکیل پیوندهای آهن-اکسیژن بودند و در طول موج cm-1 575 پیکهای متفاوتی با نمونههای شاهد مشاهده شد.

کاربرد منطق فازی و سیستم استنتاج تطبیقی عصبی- فازی جهت پیشبینی تغییرات فیزیکی و شیمیایی و طبقهبندی کیفی لیموشیرین پوششدار طی مدت نگهداری
در پژوهش حاضر با بهرهگیری از منطق فازی و با کمک روش حداکثری-حداقلی ممدانی و یکی از توابع عضویت مثلثی، گوسی و ذوزنقهای برای طبقهبندی کیفی لیموشیرین پوششدهیشده با بهرهگیری از دو الگوریتم، یکی با پنج ورودی (سفتی بافت، مواد جامد محلول، درصد رنگ سبز، حجم و رنگ پوست) و دیگری با سه ورودی حاصل از تصویر (درصد رنگ سبز، حجم و رنگ پوست) استفاده گردید. برای پیشبینی خصوصیات کیفی (سفتی و شاخص رسیدگی) نیز از سیستم ممدانی و تــوابع عضـویت مطلوب مثلثی، ذوزنقهای، زنگولهای و گوسی با کمک سه ورودی (یعنی زمان نگهداری، زاویه رنگ و حجم) استفاده شد. نتایج نشان داد که میانگین دقت الگوریتم طبقهبندی برای توابع عضویت گوسی، مثلثی و ذوزنقهای بهترتیب 975/0، 931/0 و 960/0 بود. نکته مهم دیگر این که مدل بر مبنای شاخصهای استخراجی از تصویر نیز عملکرد بسیار خوبی داشت (صحت بالاتر از 966/0). مشخص شد که بهترین پیشبینی برای شاخص رسیدگی و سفتی بافت بهترتیب با مدل منطق فازی با توابع عضویت مثلثی (ضریب تبیین برابر 9996/0) و گوسی (ضریب تبیین برابر 9992/0) قابلدستیابی است. نتایج حاصل از تجزیه و تحلیل میزان حساسیت نشان داد که زمان نگهداری بیشترین تأثیر را هم بر شاخص رسیدگی و هم بر سفتی سطحی لیموشیرین دارد. درنهایت میتوان گفت که سیستم استنتاج عصبی-فازی عملکرد قابلقبولی در طبقهبندی کیفی و پیشبینی خصوصیات فیزیکی و شیمیایی لیموشیرین برخوردار است و با توجه به استفاده از خصیصههای استخراجی از تصاویر، بهعنوان روش غیرمخرب در سردخانهها قابلاستفاده است.

در این پژوهش جانشینی شورتنینگ تجاری در سطحهای صفر تا100 درصد با الئوژل بهینهشده بر پایه ترکیب اتیل سلولز/اسید استئاریک/استئاریل الکل/سوربیتان مونو استئارات و مخلوط روغن کانولا/سویا بر ویژگیهای کیک روغنی موردبررسی قرار گرفت. برای این منظور، از فرمولاسیون بهینهشده برای تولید الئوژل با بیشترین مشابهت با ساختار فیزیکی شورتنینگ استفاده گردید. نتایج نشان داد که تفاوت معنیداری در ویژگیهای رطوبت، فعالیت آبی و حجم مخصوص کیک تهیهشده با شورتنینگ الئوژل با کیک تهیهشده با شورتنینگ تجاری مشاهده نمیشود. از جهت رنگ پوسته، در شاخصهای روشنایی و قرمز-سبز تفاوت معنیداری مشاهده نشد و تنها در شاخص زرد-آبی، سطح 100 درصد جانشینی، مقدار کمتری نسبت به شاهد نشان داد. در شاخص تخلخل بافت، تفاوت معنیداری بین پنج نمونه موردبررسی مشاهده نشد. بررسی بافت نمونههای کیک نشان داد که سختی بافت و شاخص فنریت با افزایش درصد جانشینی شورتنینگ تجاری کاهش مییابد ولی شاخص پیوستگی تفاوت معنیداری پیدا نکرد. همچنین با افزایش درصد جانشینی شورتنینگ تجاری، با نرمتر شدن بافت، انرژی جویدن نمونه نیز کاهش یافت. درنهایت، ارزیابی حسی نمونههای کیک، تفاوت معنیداری در شاخصهای طعم، بافت، ظاهر، رنگ و پذیرش کلی نشان نداد. بهطورکلی استفاده از شورتنینگ الئوژل بهجای شورتنینگ تجاری در فرمولاسیون کیک، نهتنها در سطح 50 درصد جانشینی، میتواند کیک با ویژگیهای قابلقبول ای بههمراه دارد، حتی تا سطح 100 درصد نیز میتواند جایگزین شورتنینگ تجاری گردد و تنها در شاخص بافت تا حدی بافت نرمتری را ایجاد میکند.

تهیه سامانه امولسیون ژل سرد با استفاده از پیکرینگ های کمپلکس ایزوله پروتئین سویا-صمغ دانه ریحان بهعنوان جایگزین چربی در خامه
مصرف زیاد چربی خطر ابتلا به بیماریهای مختلفی را افزایش می دهد، اما بهدلیل نقش چندگانه چربی در فرآوردههای غذایی، حذف یا کاهش آن منجر به ایجاد خصوصیات کیفی نامطلوب میگردد. بنابراین، در این تحقیق استفاده از ساختار امولسیونژل سرد پایدارشده با استفاده از پیکرینگ های ایزوله پروتئین سویا (ISP) و کمپلکس ایزوله پروتئین سویا-صمغ دانه ریحان (ISP-BSG) بهعنوان جایگزین چربی موردمطالعه قرارگرفت. ذرات پیکرینگ ISP-BSG بهترتیب با نسبتهای مختلف جرمی ISP:BSG، 0:1، 1:1، 1:2 و 1:3 تهیه و بهترتیب 1S:0B ،1S:1B ،2S:1B و 3S:1B نامگذاری شدند. بررسی ویژگی های امولسیون ژل نشان داد که پوشش دهی سطح قطرات امولسیون توسط نانوذرات جامد S:B یک پوسته سفت و سخت را تشکیل میدهد. با افزودن کلسیم کلرید به امولسیون، شبکههای امولسیون ژل از قطرات روغن تجمعیافته تشکیل شدند که این ساختار در نمونههای 1S:0B و 2S:1B قابلمشاهدهتر بود. همچنین، نوع و ساختار پیکرینگ بهکاررفته بر مقدار حفظ روغن در امولسیون ژل اثرگذار بود. حضور BSG در نسبت مناسب (2S:1B) با ISP باعث ایجاد امولسیون ژل باثباتتر با استحکام مکانیکی (سختی و قوام) بالاتری شد. با توجه به پایداری و خصوصیات بافتی از بین نمونههای امولسیون ژل، دو فرمول 1S:0B و 2S:1B برای تولید خامه با چربی کاهشیافته (5، 10 و 15%) استفاده شد. استفاده از درصدهای بیشتر از 5 درصد جایگزینی با افزایش آباندازی خامه همراه بود (05/0p <)، اما نوع امولسیون ژل بر میزان آباندازی تأثیر معنیداری نداشت. بیشترین میزان پذیرش کلی با جایگزینی امولسیون ژل 2S:1B با کاهش 5% چربی حاصل شد (05/0p>).

بررسی خواص فیزیکوشیمیایی موسیلاژ بامیه و مقایسه کارآیی استخراج دو روش حلال و فوق بحرانی
خواص درمانی بامیه از پلیساکاریدهای موجود در غلاف میباشد که قسمت اعظم آن در بخش لزجی گیاه وجود دارد و موسیلاژ خوانده میشود. بخش اعظم موسیلاژ از کربوهیدراتها تشکیل شده است و خاصیت اصلی آن در مواجه شدن با مولکولهای آب است که بهصورت ژل در میآید و در صنایع مختلف کاربرد دارد. موسیلاژ در صنایع غذایی بهعنوان پایدارکننده و امولسیفایر کاربرد دارد و در صنایع داروسازی برای کنترل آزادسازی قرصها و تولید شربت استفاده میشود. هدف از این تحقیق بهینهسازی شرایط استخراج موسیلاژ از بامیه به دو روش حلال و روش فوق بحرانی جهت یافتن نقاط بهینه استخراج میباشد. بهمنظور بهدست آوردن مدل ریاضی و یافتن بیشترین تأثیر پارامترها بر پاسخ از روش سطح پاسخ استفاده شده است. در روش استخراج با حلال متغیرهای دما، زمان و نسبت حلال به ماده جامد برای رسیدن به حداکثر میزان بازده، بهترتیب ˚C 60، min 224 و 48 بهدست آمدند و دما و نسبت حلال به ماده جامد بهترتیب بیشترین تأثیر را بر بازده استخراج داشت. در روش استخراج فوق بحرانی دما، فشار و زمان بهعنوان متغیرهای مستقل در نظر گرفته شدند و برای رسیدن به حداکثر بازده بهترتیب ˚C 50، bar 5/25 و min 135 بهدست آمد. میزان بازده موسیلاژ استخراجی در نقطه بهینه در روش استخراج با حلال 42/5 و در روش فوق بحرانی 69/1 درصد بود. درصد بازده استخراج موسیلاژ در روش فوق بحرانی کمتر از روش خیساندن شد، اما میزان حلال مصرفی و زمان استخراج در روش خیساندن بیشتر از روش فوق بحرانی بود. عصاره حاصل از روش فوق بحرانی توسط GC-MS آنالیز شد و نتیجه حاکی از حضور پلیساکاریدها در نمونه میباشد.

بهینهسازی شرایط تولید ژله با استفاده از پکتین استخراجشده از پوست سیبزمینی و بررسی خواص بافتی، فیزیکوشیمیایی و حسی آن در مقایسه با پکتینهای تجاری
پکتین مخلوط پیچیدهای از پلیساکاریدها است که بهفراوانی در صنعت غذا بهعنوان ژلکننده، ثباتدهنده و امولسیفایر مورداستفاده قرار میگیرد. پوست سیبزمینی منبع غنی از پکتین با درجه استفریفیکاسیون پایین است که عموماً بهعنوان ضایعات دور ریخته میشود. برای تشکیل ژل توسط پکتینهای با درجه استری کم، غلظت پکتین، کلرور کلسیم و میزان pH بسیار مؤثر است. لذا هدف از این پژوهش بهینهسازی خواص بافتی ژله حاوی غلظتهای مختلف پکتین (5/0 و 1 درصد)، کلرور کلسیم (15، 30 و 45) و ) pH5/2 و 4) و مقایسه خواص فیزیکوشیمیایی، بافتی و حسی ژله تولیدشده در شرایط بهینه با ژله حاصل از پکتین تجاری مرکبات و سیب بود. مطابق با نتایج شرایط بهینه برای تولید ژله با استفاده از پکتین استخراجشده از پوست سیبزمینی با هدف دستیابی به بالاترین میزان سختی ژل (0959/30 نیوتن) و نیروی لازم برای شکستن ژل (3431/27 نیوتن) بهصورت همزمان با 33/94 درصد مطلوبیت در شرایط pH برابر با 4، میزان کلرور کلسیم 2286/35 میلیگرم/گرم پکتین و غلظت پکتین 1 درصد مشاهده گردید. شرایط بهینه تولید ژله حاوی پکتین پوست سیبزمینی برای ژلههای تجاری حاوی پکتین مرکبات و سیب اعمال گردید و خواص فیزیکوشیمیایی، بافتی و حسی ژلهها با یکدیگر مقایسه گردید. نتایج نشان داد اختلاف معنیداری بین خواص فیزیکوشیمیایی، بافتی و اورگانولپتیکی ژلههای تهیهشده با پکتین پوست سیبزمینی با ژلههای تجاری مرکبات و سیب مشاهده نگردید. نتایج تحقیق حاضر نشان داد با استفاده از بهینهسازی شرایط تولید میتوان از پکتین پوست سیبزمینی در فرمولاسیون ژله استفاه نمود و ژلهای با خواص کیفی مطلوب و قابل مقایسه با پکتینهای تجاری متداول تولید نمود.

ریزپوشانی پروتئین هیدرولیزشدهی گرده گل با کنستانتره پروتئین آبپنیر و فایبرزول و بررسی پایداری و ساختاری ریزکپسولها
ریزپوشانی پروتئینها و پپتیدها در حفظ ساختار و ویژگیهای عملکردی آنها تأثیرگذار است. در این پژوهش بهمنظور حفظ ساختار و پایداری پروتئین هیدرولیزشده گرده گل در مقابل عوامل تخریبکننده، ریزپوشانی پروتئین هیدرولیزشده توسط خشککن پاششی با کنستانتره پروتئین آبپنیر (WPC) و فایبرزول و مخلوط آنها انجام شد. ترکیب دیواره و پودر پروتئین هیدرولیزشده با نسبت 10 به 1 وزنی-وزنی استفاده شدند. ترکیب مورداستفاده برای دیواره شامل WPC 2 درصد، فایبرزول 2 درصد، همچنین مخلوط WPC و فایبرزول با نسبت 1 به 3 بود. بهمنظور تسریع واکنشهای اکسایشی، کپسولهای حاصله بهمدت 48 ساعت در معرض اشعه UV قرار گرفتند. بیشترین میزان مهارکنندگی رادیکال (DPPH) در مدت در معرض قرار گیری اشعه UV مربوط به کپسول با دیواره مخلوط فایبرزول و WPC بود. نتایج طیفسنجی (FTIR) نشان داد دیواره ترکیبی فایبرزول و WPC بهترین عملکرد را در حفظ ساختار شیمیایی کپسولها داشته است. تصاویر حاصل از میکروسکوپ الکترونی رویشی (SEM) نشان داد ریزکپسولهایی که دارای دیواره مخلوط فایبرزول و WPC بودند، دیواره یکنواخت و صافتری نسبت به ریزکپسولهای با دیواره فایبروزلی بودند. درنهایت مخلوط WPC و فایبرزول بهعنوان بهترین دیواره با قابلیت محافظتی مناسب بهمنظور ریزپوشانی پروتئینهای هیدرولیزشده و محافظت از آنها در مقابل عوامل تخریبکننده انتخاب گردید.
