چکیده

در این پژوهش پس از جداسازی آغازگر لاکتوباسیلوس غالب موجود در خمیرترش سنّتی حاصل از آرد کامل گندم و تائید شناسایی آن با استفاده از PCR دارای پرایمر اختصاصی، از آغازگر مذکور جهت تولید خمیرترش استفاده شد. سپس تاثیر زمان تخمیر (8 ،16، 24 ساعت) و مقدار شکر (5/0، 1، 5/1درصد) بر فعالیت آغازگر لاکتوباسیلوس جدا شده مورد ارزیابی قرار گرفت. پس از فرآوری نان قالبی با استفاده از تیمارهای خمیرترش ، بیاتی نان‌های تولیدی در یک بازه زمانی چهار روزه (2، 48 و 96 ساعت پس از پخت) براساس شاخص سفتی بافت (به روش بافت‌سنجی) مورد بررسی قرارگرفت. همچنین میزان تخلخل (به روش پردازش تصویر) و ویژگی های حسّی نان تازه خوری نیز با نمونه شاهد مقایسه شد. نتایج حاصل نشان داد که کمترین و بیشترین میزان سفتی بافت نان پس از 96 ساعت به ترتیب مربوط به نمونه های فرآوری شده با خمیرترش تخمیر شده طی 24 ساعت و دارای 5/0 درصد شکر و همچنین خمیرترش تخمیر شده طی 8 ساعت و دارای 5/1درصد شکر بود. علاوه بر این، بیشترین مقدار تخلخل نان تازه‌خوری در نمونه حاصل از تخمیر 24 ساعت خمیرترش و محتوی 5/1 درصد شکر و کمترین مقدار آن نیز در نمونه حاصل از 8 ساعت تخمیر با محتوی 5/0 درصد شکر مشاهده گردید. پس از ارزیابی نتایج حاصل از این آزمون ها، رابطه بین عوامل موثر بر تخمیر خمیرترش به‌منظور بررسی خصوصیات بافتی نان های تولیدی بر اساس رفتار ویسکوالاستیک آن ها نیز مورد بررسی قرار گرفت.
